چکیده

We use an instrument combining optical (surface plasmon resonance) and acoustic (Love mode acoustic wave device) real-time measurements on a same surface for the identification of water content in collagen and fibrinogen protein layers. After calibration of the surface acoustic wave device sensitivity by copper electrodeposition, the bound mass and its physical properties – density and optical index – are extracted from the complementary measurement techniques and lead to thickness and water ratio values compatible with the observed signal shifts. Such results are especially usefully for protein layers with a high water content as shown here for collagen on an hydrophobic surface. We obtain the following results: collagen layers include 70±20% water and are 16±3 to 19±3 nm thick for bulk concentrations ranging from 30 to 300 μg/ml. Fibrinogen layers include 50±10% water for layer thicknesses in the 6±1.5 to 13±2 nm range when the bulk concentration is in the 46 to 460 μg/ml range.

In situ evaluation of density, viscosity, and thickness of adsorbed soft layers by combined surface acoustic wave and surface plasmon resonance.

We show the theoretical and experimental combination of acoustic and optical methods for the in situ quantitative evaluation of the density, the viscosity, and the thickness of soft layers adsorbed on chemically tailored metal surfaces. For the highest sensitivity and an operation in liquids, a Love mode surface acoustic wave (SAW) sensor with a hydrophobized gold-coated sensing area is the aco...

Tunable Plasmonic Nanoparticles Based on Prolate Spheroids

Metallic nanoparticles can exhibit very large optical extinction in the visible spectrum due to localized surface plasmon resonance. Spherical plasmonic nanoparticles have been the subject of numerous studies in recent years due to the fact that the scattering response of spheres can be analytically evaluated using Mie theory. However a major disadvantage of metallic spherical nanoparticles is ...

Human immunoglobulin adsorption investigated by means of quartz crystal microbalance dissipation, atomic force microscopy, surface acoustic wave, and surface plasmon resonance techniques.

Time-resolved adsorption behavior of a human immunoglobin G (hIgG) protein on a hydrophobized gold surface is investigated using multitechniques: quartz crystal microbalance/dissipation (QCM-D) technique; combined surface plasmon resonance (SPR) and Love mode surface acoustic wave (SAW) technique; combined QCM-D and atomic force microscopy (AFM) technique. The adsorbed hIgG forms interfacial st...

Fabrication and Characterization of the Fiber Optical Taper for a Surface Plasmon Resonance Sensor

For a fiber optical surface plasmon resonance (SPR) sensor a short part of its cladding should be removed to coat a thin layer of a metal. Usually this is problematic when an optical fiber with small core diameter is used. In this paper, a new method using µliter droplet of the HF acid for short fiber optical taper fabrication is reported. Using this method in a multi-mode optical fiber w...

Simultaneous surface acoustic wave and surface plasmon resonance measurements: electrodeposition and biological interactions monitoring

Abstract We present results from an instrument combining surface acoustic wave (SAW) propagation and surface plasmon resonance (SPR) measurements. The objective is to use two independent methods, the former based on adsorbed mass change measurements and the latter on surface dielectric properties variations, to identify physical properties of protein layers, and more specifically their water co...
